The DF-1 cell line is spontaneously immortalized, can multiply indefinitely, and has a cellular morphology typical of fibroblasts.The DF-1 cells are reverse transcriptase-negative, free of endogenous gene sequences from avian sarcoma and leukemia viruses, and are non-tumorigenic.The DF-1 cell line is a globally commercialized cell line licensed by the U.S. Food and Drug Administration (FDA).
It is now widely used in the proliferation of avian viruses, expression of recombinant proteins, research on oncolytic viruses, and production of animal and human vaccines.DF-1 cells are susceptible to chicken infectious bursal virus, echinoid virus, avian leukemia virus, Laurasia sarcoma virus, and turkey herpesvirus, and can replicate on DF-1 cells, and they can form obvious cytopathic lesions.
This cell line is an adherent-dependent cell, which is normally cultured using basal medium supplemented with fetal bovine serum, with D/F1:1 + 10% fetal bovine serum being optimal. After optimization screening, an AEM medium suitable for DF-1 cell culture was identified. The medium was chemically defined and only required the addition of 3% neonatal bovine serum to maintain DF-1 cell growth. The medium supports serum-free inoculation of viruses and can be used as a serum-free virus maintenance solution to support the proliferation of certain viruses.
